DESCRIPTION:Efforts to repurpose CRISPR-Cas systems have produced a suite
of genome editing tools\, including programmable nucleases\, base editors
\, and prime editors. These tools have greatly enabled the study of genom
es and gene function\, and their advancement to therapeutic development h
as demonstrated promise for addressing a host of unmet medical needs. Our
understanding of how endogenous cellular processes influence the activit
y of these tools\, however\, lags far behind their application and\, due
to the rapid pace of technology development\, behind efforts to build new
approaches. Our work focuses on identifying cellular determinants of gen
ome editing tools to better understand how they work. Our results provide
key insights into how those tools interact with the cellular environment
and suggest general strategies for improvement.

DESCRIPTION:The annual McGinnis Memorial Lecture was established by the st
aff and students of the Departments of Microbiology and Immunology in 197
9 to honor the memory of James William McGinnis\, Jr.\, a beloved Duke do
ctoral candidate working in Bill Joklik's laboratory who died unexpectedl
y in a canoeing accident.\n\nAdolfo García-Sastre\, PhD\, is Professor of
Medicine and Microbiology and Co-director of the Global Health & Emergin
g Pathogens Institute at the Icahn School of Medicine at Mount Sinai. \n\
nThe García-Sastre Laboratory is focused on exploring virus-host interact
ions with an emphasis on virus regulation of innate and adaptive immune r
esponses. The outcome of these interactions not only determines disease s
everity but also influences the development of protective immunity result
ing from viral infection and/or vaccination. The team has developed sever
al techniques (reverse genetics)\, which allow for the genetic manipulati
on of the genomes of several virus families including influenza virus and
Newcastle disease virus. Other viruses being studied include Zika virus\
, Dengue virus\, West Nile virus\, and Crimean-Congo hemorrhagic fever vi
rus. These techniques are currently being used in several research areas
including: i) characterization of virus-encoded virulence factors\, ii) i
dentification of virus-encoded antagonists of the interferon system\, iii
) virus replication and gene expression\, iv) immune regulation of influe
nza replication\, and v) vaccine development. Reverse genetics are also b
eing utilized to generate virus vectors based on influenza virus and Newc
astle disease virus. These viruses are effective inducers of humoral and
cellular immune responses. Live attenuated influenza virus vaccines are b
eing generated by genetic modification of the influenza virus-encoded int
erferon antagonist that was originally identified by the laboratory. Also
\, recombinant viruses can be produced that express protective antigens o
f other pathogens for which no safe attenuated vaccine strains are availa
ble.

DESCRIPTION:Reception to follow I Open to the public\nFree parking in Re
search Drive Garage\n\nWe live in an age in which we cede more and more o
f life to machines\, raising questions for the practice of medicine. Wha
t is the calling of medicine? Is it not to heal human beings? But what
does it mean to heal? Is a human being just a body? Is the body just a
machine? Is a doctor just a mechanic? Can machines substitute for human
skill? Can they erode human skill? How should we think of death? What
\, after all\, is a good life? It's unlikely we would all agree on the a
nswers to these questions\, but they do all need to be asked. And underst
anding the role played by lateralization in brain function may guide us t
owards what are at least better answers to these pressing questions. \n\n

DESCRIPTION:Single-cell paired multi-omic data\, such as jointly sequenced
RNA and ATAC via 10x Multiome\, has been widely adopted to study the reg
ulatory mechanisms in different biological systems. However\, there are s
till open questions on how to broadly measure the coordination between th
ese two modalities within individual cells and account for the changing c
oordination over time. In this talk\, we discuss two different lines of w
ork with similar themes that model the RNA-ATAC coordination holistically
. In the first half of the talk regarding cortical development\, we prese
nt the Tilted-CCA\, a new statistical decomposition that uncovers the com
mon and modality-unique geometric relations among cells. This decompositi
on gives us a new perspective for studying development since a gradual in
crease and decrease of RNA-ATAC coordination can mathematically quantify
the poised chromatin state. In the second half of the talk regarding canc
er resistance\, we present a cell-wise lineage imputation score. This sta
tistical regression method uses static lineage barcoding data that our co
llaborators have developed to work with the 10x Multiome. Our method offe
rs new insight into cancer resistance mechanisms\, as our statistical met
hod can assess how many progenies each cancer cell is likely to produce a
t a future time. This method allows us to quantify how a cancer subclone'
s lineage size\, its RNA+ATAC profile\, and its plasticity contribute tow
ards said subclone's resistant behavior.

fessor of psychiatry & behavioral sciences and physician at the Durham VA
Health Care System\, shares stories and insights from his new book\, "Fa
cing the Unseen: The Struggle to Center Mental Health in Medicine\," in a
conversation with Julie Penzner\, MD\, vice chair for education in psych
iatry & behavioral sciences.\n\nIn the book\, to be published on April 9\
, Tweedy guides us through his days working in various clinical settings
as he meets people from all walks of life who are grappling with physical
and psychological illnesses. He argues for a more comprehensive and inte
grated approach\, where people with mental illness have a health care sys
tem that places their full well-being front and center.\n\nTweedy's first
book\, "Black Man in a White Coat" (2015) was a New York Times bestselle

DESCRIPTION:This talk asks: How do we relate to the work of healthcare? Ye
ars after COVID-19's emergence\, the pandemic's social and structural eff
ects on different healthcare worker cadres remain understudied. Faculty p
resenters will share material from a collaborative ethnographic study of
the lives and labors of critical care workers in a hospital intensive car
e unit (ICU) from 2020-2022. As physicians and ethnographers of medicine\
, they will highlight the moral dilemmas and structural changes that shap
