DESCRIPTION:Consolidation of declarative memories requires hippocampal-neo
 cortical communication. Although experimental evidence supports the role 
 of sharp-wave ripples in transferring hippocampal information to the neoc
 ortex\, the exact cortical destinations and the physiological mechanisms 
 of such transfer are not known. We used a conducting polymer-based confor
 mable microelectrode array (NeuroGrid) to record local field potentials a
 nd neural spiking across the dorsal cortical surface of the rat brain\, c
 ombined with silicon probe recordings in the hippocampus\, to identify ca
 ndidate physiological patterns.\n\nParietal\, midline\, and prefrontal\, 
 but not primary cortical areas\, displayed localized ripple (100 to 150 h
 ertz) oscillations during sleep\, concurrent with hippocampal ripples. Co
 upling between hippocampal and neocortical ripples was strengthened durin
 g sleep following learning. These findings suggest that ripple-ripple cou
 pling supports hippocampal-association cortical transfer of memory traces
 .

DESCRIPTION:The reversible polymerization of proteins is a process necessa
 ry for life. In neurodegeneration\, the abnormal and seemingly irreversib
 le polymerization of several different abundant proteins in the brain pat
 hologically defines disease into predictable stages. Linkage analyses and
  genome-wide associations prove that these proteins that form fibrils\, o
 ligomers\, and other conformations are not mere bystanders in disease sus
 ceptibility but can drive neurodegeneration. Yet\, inclusion abundance of
 ten poorly predicts clinical phenotypes\, and mixed pathologies common in
  patients that involve several different proteins challenge therapeutics 
 that directly target protein aggregation. Recent studies have nominated c
 ritical modulators of disease susceptibility and progression that include
  inflammation\, microbiome\, and the cell-to-cell propagation of abnormal
  proteins. Biomarkers that unravel disease heterogeneity to identify pati
 ents that might benefit most from targeting these critical modifiers prom
 ise to revolutionize bench-to-clinic approaches. With better precision in
  therapeutic strategies\, it is hoped that the first wave of disease modi
 fying drugs is already under development\, with many more promising avenu
 es yet to be explored.

DESCRIPTION:The olfactory epithelium in the nose is the peripheral organ f
 or the sense of smell\, housing primary olfactory sensory neurons. Olfact
 ory neurons are generally felt to be short-lived\, requiring ongoing repl
 acement from basal stem cells to maintain olfactory function. Anosmia (a 
 loss of the sense of smell) in humans can occur in neurodegenerative dise
 ases or due to inflammation\, tissue damage or aging. Evidence suggests t
 hat neurogenic exhaustion\, or excess cumulative damage\, may contribute 
 to olfactory loss. Accordingly\, our work has focused on understanding me
 chanisms involved in ongoing adult olfactory tissue maintenance. The lab 
 has used mouse models of olfactory injury and culture systems to study ad
 ult olfactory degeneration and regeneration. Findings from these approach
 es have led to a focus on the role of Polycomb complexes in the epigeneti
 c regulation of olfactory cellular renewal. In addition\, we have explore
 d the culture expansion and therapeutic potential of neurogenic olfactory
  basal progenitor cells.

DESCRIPTION:Dr. Romani joined Janelia in the fall of 2014 to study the dyn
 amics of cortical circuits. \n\nHis experience in the field of computatio
 nal neuroscience started at the University of Rome La Sapienza. Under the
  supervision of Daniel Amit\, covering the end of his Laurea studies and 
 most of the PhD period\, he witnessed unsupervised recurrent neural netwo
 rks learn discrete sets of stimuli.\n\nHe was a postdoc with Misha Tsodyk
 s at the Weizmann Institute of Science and at the Center for Theoretical 
 Neuroscience. There he mingled attractor networks and short-term synaptic
  plasticity\, which resulted in cue-less retrieval of information from me
 mory and the dynamics of spatial coding in hippocampus-like circuits.

DESCRIPTION:Bianca Jones Marlin is a neuroscientist and postdoctoral resea
 rcher at Columbia University's Zuckerman Institute.  She holds a Ph.D. in
  neuroscience from New York University School of Medicine\, and dual bach
 elor degrees in biology and adolescent education from St. John's Universi
 ty. \n\nAs postdoctoral fellow in the laboratory of Nobel Laureate Dr. Ri
 chard Axel\, she investigates transgenerational epigenetic inheritance\, 
 or how trauma in parents affect the brain structure and sensory experienc
 e of their future offspring. During her graduate studies\, in the laborat
 ory of Dr. Robert Froemke\, Dr. Marlin examined how the brain adapts to c
 are for a newborn. Her findings uncovered a fundamental role of the neuro
 modulator oxytocin during the transition to motherhood.\n\nDr. Marlin aim
 s to utilize neurobiology and the science of learning to better inform bo
 th the scientific and educational community on how positive experiences d
 ictate brain health\, academic performance\, and social well being.

DESCRIPTION:Changes in behavioral state are associated with modulation of 
 sensory processing in mice. Recent studies have shown that locomotor acti
 vity and arousal influence both spontaneous activity and sensory-evoked r
 esponses across mouse sensory cortices\, with important consequences for 
 sensory perception. To what extent behavioral state modulation generalize
 s to other brain functions\, however\, remains unclear. \n\nIn this talk 
 she will present our work showing that behavioral state modulates perform
 ance in delay eyeblink conditioning\, a cerebellum-dependent form of Pavl
 ovian conditioning. they find that locomotor activity improves associativ
 e learning through mechanisms that are dissociable from arousal and indep
 endent of the modulation of cortical sensory responses. Further\, optogen
 etic circuit dissection suggests that locomotion modulates delay eyeblink
  conditioning through mechanisms acting on the mossy fiber pathway within
  the cerebellum.\n\nFinally\, she will show that controlling locomotor ac
 tivity rescues the impaired eyeblink conditioning of cannabinoid receptor
  knockout mice\, highlighting the importance of considering general chang
 es in behavioral state as a powerful means through which individual genes
  contribute to complex behaviors.

DESCRIPTION:Theoretical and computational approaches are rapidly changing 
 the landscape of biology\, with theoretical considerations increasingly c
 entral to how fundamental biological questions are formulated and resolve
 d. My formal education is in physics and mathematics\, and my research fo
 cuses on building bridges between the quantitative sciences and biology. 
 I began by studying molecular biophysics with Karl Freed and Tobin Sosnic
 k at the University of Chicago. I then moved to the Physics Department at
  Stanford for graduate school\, where my biophysics interests merged with
  neuroscience through research with Mark Schnitzer and Tom Clandinin. Bef
 ore coming to Janelia\, I was a Swartz Fellow in Theoretical Neuroscience
  in the Center for Brain Science at Harvard. This independent postdoctora
 l position gave me an opportunity to interact broadly with the neuroscien
 ce community at Harvard and beyond\, and during this time I was fortunate
  to work with Haim Sompolinsky\, Florian Engert\, Damon Clark (Yale)\, Ru
 ben Portugues (MPI Neurobiology)\, and many of their talented group membe
 rs. At Janelia I am continuing to combine theoretical biophysics and neur
 obiology to understand how fundamental theoretical principles can reveal 
 order in the stunning complexity of the brain.

DESCRIPTION:Ashok Litwin-Kumar is an assistant professor in the Department
  of Neuroscience at Columbia University. He received a B.S. in physics fr
 om Caltech in 2008 and a Ph.D. in computational neuroscience from Carnegi

 his postdoctoral training at the Center for Theoretical Neuroscience at C

 He has been a faculty member of the Center since 2018. His research focus
  is theoretical neuroscience\, specifically models of learning and behavi
 or. Topics include how populations of neurons represent the sensory envir
 onment and how these representations change during learning\, the constra
 ints that anatomy and physiology place on the learning algorithms impleme
 nted in the brain\, and how these algorithms compare to those commonly st
 udied in machine learning and artificial intelligence. Models are constra
 ined by data from experimental collaborators and are used to suggest new 
 experiments to test their predictions.

DESCRIPTION:Our primary scientific interest lies in understanding how the 
 brain forms percepts and uses them to make decisions\, especially in the 
 visual domain. In particular\, we are interested in how the brain's perce
 ptual beliefs about the outside world are represented by the responses of
  populations of cortical neurons and how their spiking activity gives ris
 e to percepts and decisions. To that end we construct mathematical models
  that aim to explain neural responses and behavior.

DESCRIPTION:At my lab we are interested in forming an integrated picture o
 f learning\, memory and development\, processes that share many common me
 chanisms. We do this by developing mathematical models of the substrates 
 of these processes\, and studying these models using both computer simula
 tions and analytical techniques. Since we strive to develop an integrated
  theory that can account for these processes\, our studies span many leve
 ls of description from the molecular basis of synaptic plasticity to the 
 system level properties of the visual cortex. The research in my lab is a
 lways closely related to experimental studies\, and I have often closely 
 collaborated with experimental labs.
