DESCRIPTION:Dr. Georgia L. Wiesner is a Professor of Medicine at Vanderbil
 t University Medical Center and is board-certified in both Internal Medic
 ine and Medical Genetics and has spent the majority of her career in the 
 fast evolving area of cancer genetics. She serves as the founding Directo
 r of the Clinical and Translational Hereditary Cancer Program in the Vand
 erbilt-Ingram Cancer Center\, a NCI designated comprehensive cancer cente
 r. Established in 2012\, this program is devoted to the care of patients 
 and families with an increased risk for cancer. To date\, over 5000 patie
 nts have been evaluated in this program. Dr. Wiesner's research interests
  include family based studies for colon cancer and the translation of gen
 etic tests into clinical care.  Current projects include membership in th
 e eMERGE III network\, that is sequencing and returning the results for a

DESCRIPTION:During the past decade\, a new generation of high-throughput D
 NA sequencers has transformed biomedical and biotechnology research. Thes
 e new technologies have fostered the development of a wide range of appli
 cations to basic and clinical research\, including SNP discovery\, transc
 riptome profiling\, genome sequencing\, and epigenetics. The goal of this
  introductory course is to teach the basic principles of next generation 
 sequencing technology (NGS) and to present an overview of various library
  preparations and their applications. Advantages and limitations of vario
 us methods will be discussed and compared across technologies/platforms (
 Illumina\, PacBio\, Oxford Nanopore\, Ion Torrent). This course will also
  provide an introduction to primary data analysis and data quality assess
 ment steps. Attendees will become familiar with NGS technology terms and 
 fundamentals\, NGS data format and quality\, and will acquire a better un
 derstanding of how to choose a suitable NGS sequencing method or instrume

 itative analysis of peptides\, proteins and metabolites for both basic an

DESCRIPTION:Metabolomics has emerged as a powerful approach for characteri
 zation of molecular systems and also development of biomarkers for diseas
 e progression or diagnosis. Broadly\, metabolomics is the characterizatio
 n of small molecules by mass spectrometry and can include both "unbiased"
  or non-targeted techniques\, as well as "targeted" methods.   The measur
 ement of metabolites by mass spectrometry is also directly translatable t
 o the clinic\; many common assays such as amino acids\, acylcarnitines\, 
 vitamin D epimers\, steroid hormones\, and drugs of abuse are all clinica
 l mass spec assays.  Whether developing a novel assay or using a validate
 d metabolite assay\, the most important aspect for a successful metabolom
 ics study is deciding which technique to use and understanding the data e
 ach approach will likely be able to provide. In this course\, we will dis
 cuss sample types which are amenable to metabolomics\, and utilize case s
 tudies to discuss the critical differences in targeted and non-targeted m
 etabolomics and an investigator might choose one over another.  We will u
 se example datasets to demonstrate techniques for analysis of high dimens
 ional metabolomic data.  We will also cover the methods needed for accura
 te quantification\, how to enable longitudinal translation of metabolomic
 s assays\, and how a targeted mass spec assay may differ in utilization f

DESCRIPTION:Chromatin profiling in single cells has been extremely challen
 ging and almost exclusively limited to histone proteins. In cases where s
 ingle cell methods have shown promise\, many require highly specialized e
 quipment or cell type specific protocols and are relatively low throughpu
 t. Here\, we combine the advantages of tagmentation\, linear amplificatio
 n and combinatorial indexing to produce a high throughput single cell DNA
  binding site mapping method that is simple\, inexpensive\, and capable o
 f multiplexing several independent samples per experiment. Targeted Inser
 tion of Promoters (TIP-seq) uses Tn5 fused to protein A to insert a T7 RN
 A polymerase promoter adjacent to a chromatin-bound protein of interest. 
 Linear amplification of flanking DNA with T7 polymerase provides much hig
 her starting material prior to sequencing library preparation and yields 
 >10-fold higher unique reads per single cell compared to other methods.

DESCRIPTION:Liquid chromatography coupled with tandem mass spectrometry (L
 C/MS/MS) continues to be the key technology for the qualitative and quant
 itative analysis of peptides and proteins for both basic and clinical res
 earch projects. This GCB Academy session is designed as an introduction f
 or researchers needing to expand their knowledge of the use of LC/MS/MS-b
 ased methods for proteomics\, and thus help researchers better understand
  how these technologies can help inform their research goals. Background 
 material in basic protein chemistries will be provided\, with an emphasis
  on how to use the physicochemical characteristics of these biomolecules 
 for sample preparation specifically for LC/MS/MS analyses. In addition\, 
 the fundamentals of liquid chromatography and mass spectrometry will be d
 iscussed to enable students to understand the nuances of the experimental
  designs required to address their specific project. Real-world examples 
 will be used to illustrate sample preparation and analysis strategies\, i
 ncluding basic identification projects\, characterization of Post-Transla
 tional Modifications and differential expression analyses (including 'omi
 c biomarker discovery and targeted biomarker verification). Finally\, the
  use of open source software tools for interpretation of these datasets w

 design\, focusing on proteomic analysis of protein post-translational mod
 ifications (PTMs) and protein expression in (but not limited to) mammalia
