DESCRIPTION:Polyunsaturated fatty acids undergo enzymatic oxidation and ge
 nerate diverse lipid mediators that play crucial roles in regulating immu
 ne cell behavior\, blood clotting\, blood vessel permeability\, and tissu
 e repair. This seminar will discuss the significance of polyunsaturated f
 atty acid metabolism in relation to the cardiopulmonary effects of air po
 llution within real-world populations.\n\nDr. Yan Lin is a postdoctoral a
 ssociate at Duke Global Health Institute. He received his Ph.D. degree fr
 om UCLA in 2019. Dr. Lin's research interests revolve around investigatin
 g the health effects of air pollution and climate change. His research wa
 s built upon efforts to (1) discover novel exposure biomarkers that are m
 ore sensitive and specific to emerging pollution sources (e.g.\, wildfire
 )\, and (2) identify key signaling pathways that mediate the health effec
 ts of air pollution and/or climate change. These dual efforts contribute 
 synergistically\, ultimately facilitating the identification of environme
 ntal etiological factors of cardiopulmonary disease and adverse birth out

DESCRIPTION:RAS genes are commonly mutated in human cancer\, which is well
  established to initiate and promote tumorigenesis.  Despite there being 
 over 50 possible such 'oncogenic' mutations in these genes\, each cancer 
 type has a tropism towards a specific and often unique subset of these mu
 tations.  As RAS mutations typically occur early during tumorigenesis\, i
 f not being the initiating mutation\, these mutational patterns ostensibl
 y reflect fundamental biology underlying the process of tumor initiation.
   Thus\, elucidating the mechanisms behind RAS mutation tropism will prov
 ide insight into one of the most foundational questions in cancer biology
  - how cancer originates - which has predictive and even preventative cli
 nical implications. To determine how these patterns are established\, the
  initiating mutagenic event has to be captured in normal cells in vivo at
  the moment they occur.  It is challenging\, if not impossible\, to ident
 ify a spontaneous mutagenic event in one RAS gene in a single specific no
 rmal cell decades before manifesting as cancer in humans.  However\, the 
 environmental carcinogen urethane exhibits a profound specificity for pul
 monary tumors driven by a single specific RAS in mice. To elucidate the p
 rinciples underlying this RAS mutation tropism of this carcinogen\, we ad
 apted an error-corrected\, high-throughput sequencing approach to detect 
 mutations in murine Ras genes at great sensitivity. This analysis not onl
 y captured the initiating Kras mutation days after urethane exposure\, bu
 t revealed that the sequence specificity of urethane mutagenesis\, couple
 d with transcription and isoform locus\, to be major influences on the ex
 treme tropism of this carcinogen.\n\nDr. Counter is the George Barth Gell
 er Distinguished Professor of Pharmacology and the Associate Director of 
 the Duke Cancer Institute. His laboratory studies the origins of human ca
 ncers.
