DESCRIPTION:Single-particle tracking methods have shifted the landscape of
  physical virology from bulk methods requiring ensemble averaging to the 
 single virus level\, yielding new quantitative information about the viru
 s infection process. However\, existing methods are unable to study the c
 ritical extracellular phase of this process. In this phase\, where the vi
 rus diffuses to meet the cell surface\, the disparate spatial scales betw
 een viruses and cells\, combined with the rapid motion of viruses makes s
 tudying their interaction speed an impossible task for any single microsc
 ope. \nHere we present 3D Tracking and Imaging microscopy (3D-TrIm): a mu
 lti-modal solution that simultaneously acquires co-registered volumes fea
 turing the trajectories of fluorescent virus-like particles (VSV-G pseudo
 typed lentivirus) and two-photon volumetric images of the surrounding liv
 e-cell environment. This approach yields an unprecedented 3D view into th
 e interaction of virions at the cellular interface. Using this method we 
 tracked virus-like particles both inside and outside of the cell\, from f
 irst contacts with the cell membrane to intracellular trafficking and int
 o multi-layer epithelial tissue.

DESCRIPTION:We have developed a single-photon light-sheet imaging system t
 hat allows us to measure activity of hundreds of retinal neurons at speci
 fic laminar depths\, while simultaneously driving their activity though p
 atterned visual stimulation (Fig. 1a). We have validated the performance 
 of our system by imaging retinas from mice expressing GCaMP6f in subpopul
 ations of BCs and RGCs (Fig. 1c\, d). We measured calcium fluorescence in
  hundreds of RGCs across a field of view extending ~1.5mm each dimension\
 , under spontaneous and visual stimulus driven conditions\, at single cel
 l resolution for extended periods of time. A semi-automated algorithm was
  used for extracting the responses of RGCs to different stimuli such as m
 oving bars\, checkerboard noise and contrast and luminance modulation\, a
 nd then classifying RGCs into distinct functional types. We were also abl
 e to extract spatiotemporally precise calcium responses in dendrites\, es
 pecially dendritic spines of individual RGCs. Finally\, we were able to m
 easure fluorescence changes caused by influx of calcium in the synaptic t
 erminals of different BCs that contact RGCs and characterize the fluoresc
 ence responses based on their spatiotemporal correlations. Thus\, this pl
 atform enables characterization of neural circuit functions of distinct p
 opulations of neurons in specific layers of the retina. Our next step is 
 to combine this technology with a multi-electrode array\, to achieve a hi
 gh-throughput characterization of neural circuit computations in synaptic
 ally coupled BC-RGC circuits and their impact on the retinal output.

DESCRIPTION:As the most abundant renewable energy source\, solar energy is
  a crucial component of decarbonizing the global energy sector. However\,
  the widespread adoption of solar energy could intensify land competition
  among activities\, leading to potential environmental impacts\, such as 
 biodiversity loss. This land use challenge is due in part to the inherent
  limitations of market-dominating silicon photovoltaics\, which have limi
 ted flexibility and low theoretical efficiency (~29%).\nHybrid materials\
 , such as metal-halide perovskites\, offer promising solutions for advanc
 ing photovoltaics by combining desirable optical\, electronic\, and struc
 tural properties beyond those of silicon. In this talk\, I will first det
 ail my work on the design of perovskite heterostructures to advance next-
 generation photovoltaics in two emerging directions: luminescent solar co
 ncentrators (LSCs) for land-saving integrated photovoltaics\, and high-ef
 ficiency tandem photovoltaics. For LSCs\, I introduce the concept of narr
 owband exciton routing using phase-tuned self-assembled quantum wells\, w
 hich allows for low-loss perovskite-based LSCs with high internal quantum
  efficiency and large dimensions. For tandem solar cells\, I develop mixe
 d-dimensional heterojunctions to reduce the oxidation of tin-lead perovsk
 ite bottom cells\, ultimately leading to over 26%-efficiency all-perovski
 te tandem solar cells - exceeding the efficiency of single-junction perov
 skites for the first time.\nI will then describe the commercialization pr
 ospects of next-generation photovoltaic technologies. My recent research 
 has shown that interface engineering can enhance the performance of perov
 skite solar cells\, but at the expense of high-temperature operating stab
 ility. Through the design of molecular passivators\, I have demonstrated 
 commercial-viable operating-stable single-junction perovskites. However\,
  the commercialization of high-efficiency tandem/triple-junction technolo
 gies require more innovative solutions for stability breakthroughs. Furth
 ermore\, sustainability challenges must be addressed to ensure the large-
 scale deployment of these technologies. This includes replacing toxic sol
 vents\, minimizing lead leakage and end-life recycling strategies.

DESCRIPTION:Abstract: During cardiovascular development\, peristaltic cont
 raction of the embryonic heart tube produces time-varying hemodynamic for
 ces and pressure gradients across the atrioventricular canal. However\, t
 he relative importance of myocardial contraction and hemodynamic force to
  modulate cardiovascular morphogenesis in the non-Newtonian flow regime r
 emains poorly understood. By developing the 4-D light-sheet fluorescent m
 icroscope and post-imaging machine-learning algorithms\, we recapitulate 
 flow-mediated molecular transducers underlying the vascular injury and re
 generation in the zebrafish vascular injury model. We further elucidate e
 xercise training-mediated transcriptomic interaction underlying activatio
 n of endothelial SCD1 to catalyze anti-inflammatory metabolites. In this 
 context\, we integrate multi-scale mechano-transcriptomic interaction fro
 m zebrafish to mouse models and demonstrate flow-responsive molecular tra
 nsducers as therapeutic targets for cardiovascular development\, repair\,
  and protection.
