  know you can create next-level light effects and also engineer your own

 s Agency (DARPA) has held to a singular and enduring mission: to make piv
 otal investments in breakthrough technologies for national security. The 
 genesis of that mission and of DARPA itself dates to the launch of Sputni
 k in 1957 and a commitment by the United States that\, from that time for
 ward\, it would be the initiator and not the victim of strategic technolo
 gical surprises. This talk will discuss the mission of DARPA as it applie
 s to warfighter protection. Progress on programs designing materials\, de
 vices\, and cell-based systems to reduce threats and maximize warfighter 
 readiness will be presented. Specific examples of biotechnologies and dev
 ices that can operate within the gastrointestinal tract will be highlight
 ed.

DESCRIPTION:Glioblastoma (GBM) is an aggressive form of primary brain canc
 er with a dismal prognosis despite therapeutic intervention. With standar
 d of care\, patients with GBM have a median survival of less than one yea
 r with only 2% of patients surviving to three years. With this poor progn
 osis\, there is a desperate need for new therapies for this fatal disease
 . Interstitial therapy\, delivered at the time of surgical resection\, ca
 n be a transformational option to deliver chemotherapy or immunotherapy. 
 For this purpose we use acetalated dextran (Ace-DEX) nanofibrous scaffold
 s which have important advantages over the current FDA approved interstit
 ial therapy (Gliadel). Ace-DEX scaffolds have uniquely tunable degradatio
 n rates that significantly impact therapeutic efficacy and they offer fac
 ile scalability between pre-clinical and clinical models. To explore this
  therapy in a clinically relevant model\, we used an image guided mouse m
 odel of GBM resection and recurrence. Our studies show that Ace-DEX scaff
 olds degrade across a wide range of rates to release the chemotherapeutic
  paclitaxel  from 7 days to more than 35 days\, based on the cyclic aceta
 l coverage (CAC) of the polymer. The effect of release is evident with a 
 78% long term survival observed for mice treated with combined fast and s
 low-release scaffolds\, in comparison to a survival of 20% when the same 
 dose is delivered at a medium release rate. By incorporating the immune a
 djuvant resiquimod into the scaffold instead of chemotherapy\, we show th
 at Ace-DEX scaffolds can modulate the immunosuppressive environment of th
 e cancer to enhance clearance of the tumor and illustrate long-term memor
 y and protection against subsequent tumor challenges out past 100 days af
 ter initial tumor inoculation. Our preclinical results indicate that we c
 an enhance delivery of chemotherapeutics or immunotherapy with the applic
 ation of Ace-DEX nanofibrous scaffolds to develop translational therapies
  to better treat this devastating cancer.

DESCRIPTION:Understanding Surprises in Nonlinear Microscopy: Unexpected Co
 mplexity Creates Useful Contrast\n\nNonlinear microscopy can detect molec
 ular signatures that are fundamentally different (and in many cases more 
 useful) than what is found using conventional microscopes. Our lab has ma
 de technological advances that allow excellent signals with very low aver
 age laser powers (far less than a laser pointer) that avoid damage\, even
  in very sensitive samples.  Here we will focus on two specific applicati
 ons that demand such care: detecting molecular anomalies in melanoma spec
 imens that indicate metastatic cancer before it is clinically obvious\, a
 nd understanding historically important pigments in cultural heritage obj
 ects.  In both cases\, the remarkable complexity in the nonlinear pump-pr
 obe signals\, due to many different mechanisms\, makes it possible to dis
 tinguish chromophores that look identical by traditional means. I will di
 scuss new approaches that separate the different signal contributions by 
 physical mechanism\, thus better separating useful contrast from backgrou
 nd.  This has the potential to significantly reduce melanoma mortality\, 
 and the potential to detect early signs of degradation in artwork before 
 they manifest in visual distortions.

DESCRIPTION:Hydrogels\, highly hydrated cross-linked polymer networks\, ha
 ve emerged as powerful synthetic analogs of extracellular matrices for ba
 sic cell studies as well as promising biomaterials for regenerative medic
 ine applications. A critical advantage of these synthetic matrices over n
 atural networks is that bioactive functionalities\, such as cell adhesive
  sequences and growth factors\, can be incorporated in precise densities 
 while the substrate mechanical properties are independently controlled. W
 e have engineered poly(ethylene glycol) [PEG]-maleimide hydrogels for loc
 al delivery of therapeutic proteins and cells in several regenerative med
 icine applications. For example\, synthetic hydrogels with optimal bioche
 mical and biophysical properties have been engineered to direct human ste
 m cell-derived intestinal organoid growth and differentiation\, and these
  biomaterials serve as injectable delivery vehicles that promote organoid
  engraftment and repair of intestinal wounds. In another application\, hy
 drogels presenting immunomodulatory proteins induce immune acceptance of 
 allogeneic pancreatic islets and reverse hyperglycemia in models of type 
 1 diabetes. Finally\, injectable hydrogels delivering anti-microbial prot
 eins eradicate bone-associated bacterial infections and support bone repa
 ir. These studies establish these biofunctional hydrogels as promising pl
 atforms for basic science studies and biomaterial carriers for cell deliv
 ery\, engraftment and enhanced tissue repair.

DESCRIPTION:Many pressing neuroscience questions have not yet been answere
 d by lack of adequate experimental capabilities. This presentation will s
 how how our laboratory currently tackles this problem by bringing togethe
 r optical engineers\, computer scientists\, and neuroscientists to develo
 p new technology tailored to the rapidly evolving needs of experimental n
 eurosciences.\n\nThis presentation will focus on our recent progress in a
 ll-optical neuromodulation techniques. Optogenetics is a recent scientifi
 c breakthrough that enables neuroscientists to activate or inhibit neural
  activity in individual neurons using light. However\, the full potential
  of optogenetic neuromodulators can only be realized when they are paired
  with equally versatile light sculpting techniques. Neuroscientists requi
 re light sculpting techniques with high spatial and temporal resolution t
 o address functionally defined neural ensembles on demand at extreme spee
 ds. These requirements are best met by computer-generated holography meth
 ods but involves time-consuming nonlinear computations for which current 
 algorithms require several seconds. Our team has recently leveraged deep 
 learning models to dramatically accelerate the computation of 3D hologram
 s\, reducing the computation times by several orders of magnitude. Additi
 onally\, by implementing sparse representations of the addressable volume
  of brain tissue\, we have further reduced computation times to only a fe
 w milliseconds\, which matches the standard time scale of neural events. 
 The discussion will focus on future applications of these technologies an
 d their continued development to best serve the broader neuroscience comm
 unity.\n\nDr. Nicolas Pégard is an assistant professor in the departments
  of Applied Physical Sciences and Biomedical Engineering at UNC Chapel Hi
 ll\, and a primary faculty in the UNC Neuroscience Center. His research i
 n optical instrumentation aims to extend the field of microscopy beyond i
 maging techniques\, with new optical technologies tailored to probe and m
 anipulate biological systems at the cellular scale with light. His resear
 ch is supported by a 2023 Sloan research fellowship in Neurosciences\, a 
 2021 Beckman Young Investigator award\, and a 2018 Career Award at the Sc
 ientific Interface from the Burroughs Wellcome Foundation.

DESCRIPTION:Evolution occurs when selective pressures from the environment
  shape inherited variation over time. Within the laboratory\, evolution i
 s commonly used to engineer proteins and RNA\, but experimental constrain
 ts have limited our ability to reproducibly and reliably explore key fact
 ors such as population diversity\, the timing of environmental changes\, 
 and chance. We developed a high-throughput system for the analytical expl
 oration of molecular evolution using phage-based mutagenesis to evolve ma
 ny distinct classes of biomolecules simultaneously. In this talk\, I will
  describe the development of our open-source python:robot integration pla
 tform which enables us to adjust the stringency of selection in response 
 to real-time evolving activity measurements and to dissect the historical
 \, environmental\, and random factors governing biomolecular evolution. F
 inally\, I will talk about our many on-going and future projects which ut
 ilize this system to evolve previously intractable biomolecules to target
  the undruggable proteome of protein-protein interactions.

DESCRIPTION:Electro-optically tunable active metasurfaces that enable dyna
 mic modulation of reflection amplitude\, phase\, and polarization using r
 esonantly excited materials and phenomena are powerful design elements fo
 r meta-imaging and computation.  As flat\, low-profile optical elements\,
  active metasurfaces have potential serve as cascadable\, programmable co
 mponents in optical meta-imaging systems\, such as lens-less cameras and 
 single-photon imaging systems.  \nActive metasurfaces that enable dynamic
  complex index modulation to vary the amplitude\, phase and polarization 
 have been recently explored using several active materials and modulation
  phenomena\, including carrier index in plasmonic ENZ structures\, reorie
 ntation of liquid crystal molecules\, electrooptic effects in quantum wel
 l heterostructures and polar perovskite materials\, as well as index chan
 ges in phase change materials.   Recently also\, metasurfaces that employ
  dielectric phase-gradient elements with high quality factor non-local as
  well as local resonances have expanded the design space for active metas
 urfaces by enabling silicon and other more conventional  dielectric mater
 ials with modest values of index change to be utilized. \nWe can develop 
 a taxonomy for active metasurfaces based on the attainable degree of spat
 ial and temporal control. The spatial phase gradient arising from phase-r
 econfigurable array elements can enable continuous phase gradient tuning 
 for beam steering or varifocal lensing. The degree of temporal control is
  connected to the reconfiguration timescale: In quasistatic metasurfaces\
 , temporal gradients are slow compared to the period of electromagnetic w
 aves\, while time-modulated structures feature temporal gradients modulat
 ed fast enough to alter the frequency of the scattered beam. This opens t
 he possibility to frequency multiplex scattered beams from active metasur
 faces to facilitate harmonic beam steering in which a single active apert
 ure steers scattered beams for different frequencies at independent angle
 s.\nIn this talk\, I will discuss metasurfaces with high quality factor\,
  local\, resonant elements capable of two-dimensional phase gradient gene
 ration\, in both passive and active metasurface designs.  I will also des
 cribe active metasurfaces with both spatial and temporal phase gradients\
 , and an active metasurface as a lens-less imaging system\,and compare th
 e characteristics to conventional lens-coupled image sensors.

DESCRIPTION:3D bioprinting can create living human tissues on demand based
  on specifications contained in a digital file. Such highly customized\, 
 physiologically-relevant 3D human tissue models can screen potential drug
  candidates as an alternative to expensive pre-clinical animal testing. T
 he Willerth lab has developed a novel fibrin-based bioink called TissuePr
 int for bioprinting neural tissues derived from human induced pluripotent
  stem cells (hiPSCs)\, which can become any cell type found in the body. 
 Our group has bioprinted a range of functional human tissues for a variet
 y of applications\, including modeling neurodegenerative diseases like Al
 zheimer's and for screening potential cancer drugs. Here I will discuss t
 he latest work from our group as well discuss our award-winning spin-off 
 company - Axolotl Biosciences - that sells novel reagents for bioprinting
  human tissue models.

DESCRIPTION:Paul Lauterbur\, closed his seminal article on magnetic resona
 nce imaging (MRI) with the statement that "Zeugmatographic techniques sho
 uld find many useful application in studies of the internal structures\, 
 states and composition of microscopic objects." The Duke Center for In Vi
 vo Microscopy has spent the last 40 years reducing Professor Lauterbur's 
 vision to practice. This talk will describe some of the technical develop
 ments of ultra-high magnetic field\, specialized gradients\, novel rf det
 ectors and creative signal processing that have allowed us to achieve spa
 tial resolution more than 2 million times that of clinical MRI. The metho
 ds have been translated from laboratory curiosity to routine practice pro
 viding new insights into a range of mouse models of disease. We will incl
 ude applications in mouse models of normal aging and Alzheimer's disease.
  \n\nDr. G. Allan (Al) Johnson completed his BA in mathematics and physic
 s at St. Olaf College\, Northfield Minnesota in 1969. He completed his Ph
 .D. in physics at Duke University in 1974 and joined the department of ra
 diology at Duke that same year. He served as the first Director of Diagno
 stic Physics for Duke Medical Center from 1979-2009. During that period\,
  he had the good fortune to work in close collaboration with colleagues a
 t GE Medical Systems in installing the world's first high field (1.5T) cl
 inical MRI at Duke in 1983. Much of the early work was performed on dogs.
  A desire to limit studies on dogs lead Dr Johnson to ask how might MRI s
 ystems be adapted to higher spatial resolution (for mice and rats). He de
 monstrated the first in vivo images of rats at microscopic resolution in 
 1986. That same year he established The Duke Center for In Vivo Microscop
 y\, an NIH National Resource dedicated to further the development and app
 lication of MR microscopy. He and his colleagues at the CIVM have continu
 ed to push the spatial resolution of MRI through development of specializ
 ed high field (9.4T) systems with high- performance gradients and RF coil
 s and novel methods of encoding. Many of the advances are consequences of
  extensive collaboration with colleagues at Berkley\, Stanford\, Universi
 ty of Pennsylvania\, Pittsburgh University\, NIEHS and elsewhere. He is a
  Senior Fellow of the Society of Magnetic Resonance in Medicine and the A
 cademy of Radiology. He is the Charles E. Putman University Professor of 
 Radiology\, Physics and Biomedical Engineering.

DESCRIPTION:Charged\, chiral anionic semiconducting polymers helically wra
 p single-walled carbon nanontubes (SWNTs) at periodic and constant morpho
 logy (Figure 1a-b). These polymers can be to modulate SWNT electronic pro
 perties\, provide expansive solubility\, or organize functional organic m
 oieties at predefined intervals along the SWNT surface. Such helically wr
 apped polymer-SWNT superstructures\, for example\, feature perylene diimi
 de (PDI) electron acceptor unit positioned at 3 nm intervals along the na
 notube surface\, thus controlling rigorously SWNT-electron acceptor stoic
 hiometry and organization. Potentiometric studies determine driving force
 s for photoinduced charge separation (CS) and thermal charge recombinatio
 n (CR) reactions\, as well as spectroscopic signatures of SWNT hole polar
 on and PDI radical anion states. Femtosecond pump-probe transient absorpt
 ion spectroscopic experiments detail driving force- and solvent-dependent
  photoinduced CS and thermal CR dynamics\, and provide insights into the 
 factors that govern photoinduced charge transfer reactions at soft matter
 -hard matter interfaces defined by polymers and SWNTs (Figure 1c). Relate
 d systems in which a rigid polymer wraps a metallic nanotube surface modi
 fies the energy-momentum (E-k) dispersion (band structure) of the tube\, 
 driving a metallic-to-semiconductor phase transition. This strategy contr
 asts approaches that regulate electronic structural properties of bulk-ph
 ase materials which rely on altering the nature of covalent bonding. Elec
 tronic spectral\, chiro-optic\, potentiometric\, electronic device\, and 
 work function data corroborate that the magnitude of band gap opening dep
 ends on the nature of the polymer electronic structure. Polymer dewrappin
 g reverses the conducting-to-semiconducting phase transition\, restoring 
 the native metallic carbon nanotube electronic structure. Because the ene
 rgy separation between carbon nanotube valence and conduction bands depen
 ds on the polymer electronic structure\, this approach provides a new app
 roach to realize novel low band gap one-dimensional materials by design.

DESCRIPTION:HIV-1 replication is dependent on efficient incorporation of v
 iral genomes into assembling virions. RNA elements that promote genome se
 lection have been identified\, but the determinants of authentic packagin
 g fidelity and efficiency remain unknown. We recently showed that heterog
 eneous transcriptional start site usage by HIV-1 produces 5′-capped RNAs 
 beginning with one\, two\, or three 5′-guanosines (Cap1G\, Cap2G\, Cap3G\
 , respectively) that are either selected for packaging as genomes (Cap1G)
  or retained in cells as translatable mRNAs (Cap2G/Cap3G). 2H-edited NMR 
 structural studies revealed that the Cap1G transcript adopts a dimeric mu
 lti-hairpin structure that sequesters the cap\, inhibits interactions wit
 h translation initiation factor eIF4E\, and resists decapping. In contras
 t\, the Cap2G/Cap3G transcripts adopt an alternate structure with an elon
 gated central helix\, exposed splice donor residues\, and an accessible c
 ap. Extensive remodeling\, achieved at the energetic cost of a G-C base p
 air\, explains how a single 5′ guanosine modifies the function of a ~9 ki
 lobase HIV-1 transcript. Finally\, using an NMR-directed competitive pack
 aging approach\, we show that cap sequestration is an essential determina
 nt of HIV-1 genome packaging\, likely preventing cap-dependent capture by
  the cellular RNA processing and translation machinery.

DESCRIPTION:Academics across all science\, technology\, engineering\, and 
 related fields with an interest in increasing diversity and access in STE
 M are invited to hear from Dr. Michael Summers.  \n\nThe Meyerhoff Schola
 rs Program at UMBC is a nationally recognized model for increasing retent
 ion and academic performance of URM undergraduates in STEM and for prepar
 ing those undergraduates to pursue and succeed in graduate and profession
 al programs. Meyerhoff includes students of all ethnicities and backgroun
 ds who are interested in issues of diversity and inclusion in STEM. Since
  1993\, the program has graduated more than 1500 students\, of which more
  than 426 alumni have earned PhDs\, 74 have earned MD-PhDs\, 160 have ear
 ned MDs\, and 330 have earned Master's degrees (some on the way to a PhD)
 . More than 200 additional alumni are currently enrolled in STEM graduate
  and professional programs. 71 alumni currently hold faculty appointments
 . Qualified high school students who declined Meyerhoff offers and attend
 ed other universities were half as likely to graduate with a STEM degree 
 and approximately 5 times less likely to pursue or complete STEM graduate
  degrees. Despite being medium sized university with a modest population 
 of African American students (~12\,000 undergraduates\, 14% African Ameri
 can) UMBC is the top undergraduate school of origin of African American S
 TEM PhD and M.D.-Ph.D. recipients in the U.S. To determine if Meyerhoff l
 ike outcomes could be achieved at other Majority research institutions wi
 th different URM compositions\, geographies\, institutional sizes\, and c
 ultures\, but with like-minded leadership\, we established partnerships w
 ith the University of North Carolina\, Chapel Hill and Pennsylvania State
  University. Five-year outcomes indicate that the program can be adopted 
 at other institutions\, with outcomes immediately matching or exceeding t
 he Meyerhoff Program. Approaches that leverage lessons learned from succe
 ssful programs like Meyerhoff with inter-institutional partnering could s
 erve as a general paradigm for expanding inclusive excellence in STEM.
