This year marks the 50th and 35th anniversaries of the reporting of the natural products and immunosuppressants cyclosporin A (CsA; sandimmune) and FK506 (tacrolimus), respectively, and the 30th anniversary of the revelation, published in Cell in 1991, that both compounds act in a way previously not seen - as "molecular glues" that induce protein-protein associations.

On this occasion, and as the realization that additional transformative medicines also act as molecular glues continues to fuel the surge of interest in diverse chemical strategies for inducing functional protein-protein associations, I will explore the arc of this story.
